You can take it off, but if you don't put it back almost exactly from where you got it, you'll get vibration through the steering wheel at around 60-70mph.
Also the weights are really designed to be used once.

You can spray over it, mine looked ok at a distance, but when I had my wheels rebalanced the old weight was removed and the new weight needed to go on a different point, so now they look gash.
Just take them off, spray them up, take them to be rebalanced and ask them to use alloy weights that stick on the inside of the rim...

You can do both options remove it leave
Both have draw backs, removal and not refitting would cause wheel shake as above
If you paint over it when you have new tyres they will remove the old weights and that would leave you withh an untainted area

It would cost anywhere between £Free & £20 to get the wheels re-balanced after you've removed the weights and sprayed them.
But ask them to use alloy weights and not steel wheel weights, IE you want the stick on weights.

Mightaswell take all the weights off and get them rebalanced when you've done spraying them and get them put on the back, doesnt cost alot

+You tend to notice the weights at 50+.....if you don't go above said speed you won't the fact you have removed said weight (give or take) no gaurantee (sp) but your best removing them, getting them refurbed and getting a sticker to put over them (the weights on a try machine tend to work in 5g measurments)

Just mark where the weight was and put it back on? Not very hard, they just bang on
