Are there any difference on the engine itself on 1 and 3 plug ecu's?

I have bought an used engine without the loom and ecu, but I'm uncertain if there is any difference in the block and if so how can i tell it?

No the blocks themselves are physically the same.

converting a 3 plug engine to run in a single plug car does require blanking a hole off for the air injection in the cylidner head.

the exhaust on a 3 plug model has 2 lambda sensors.

I'm going to rebuild it then add a turbo.
So the exhaust will be changed, thus i don't even have it on.
Same with the intake, only have the block and cyl. head.

Are you converting the engine to one plug or 3 plug?

I dont know what the engine i have are.
So i'm trying to figure out how to see the difference, if there is one, between them.

I dont have the loom nor the exhaust manifold, so they can't give the answer.

The engine currently running in the car is a 1 plug

I can put the question this way instead.
Can i use a 3 plug TU5J4 without the manifold, intake and loom in a 1 plug car?
